The source computer for all my data is DOA. The HDD is fine and I was wondering if it was possible to use the Easy Sync software to copy the data to my new target computer. The HDD is in a USB enclosure and I was hoping to not have to find and copy the data and settings manually.

Thank you for posting your question.
Unfortunately, in order for the cable and Easy Computer Sync software to work, the drive has to be in a computer, and the Windows installation corresponding to the User files you want to transfer has to be the currently running OS for the computer.
